Shuo Huang obtained his bachelor of science degree of physics from Nanjing University in 2006. He then obtained his Ph.D degree of biophysics from Arizona State University in 2011. During 2011-2015, he was a postdoctoral fellow in University of Oxford, under the supervision of Prof. Hagan Bayley. He started his independent research career in Nanjing University in 2015. His research interests include single molecule sensing applications using engineered biological nanopores.     

Abstract:

Recent progresses in nanopore sequencing has re-defined personalized medicine and precision medicine. The commercially available MinION sequencer developed by Oxford Nanopore Technologies has already enabled direct sequencing of DNA and RNA, in a single molecule manner. However, nanopore analysis of small molecules and large structured molecules are still limited. Mycobacterium smegmatis porin A (MspA), known as the first nanopore that achieved direct sequencing of DNA, is also a high resolution sensor. Its large vestibule could enable trapping of large protein or structured RNA molecules, allowing for direct single molecule identification. When properly engineered at its pore constriction, the MspA also enables direct small molecule sensing with a high resolution. Assisted with machine learning, direct identification of nucleoside monophosphates, saccharides, alditols and amino acids were demonstrated using engineered MspA, suggesting its further use in high performance nanopore sequencing.
